RESUMEN

Artificial skins with functions such as sensing, variable stiffness, actuation, self-healing, display, adhesion, and camouflage have been developed and widely used, but artificial skins with escape function are still a research gap. In nature, every species of animal can use its innate skills and functions to escape capture. Inspired by the behavior of fish-scale geckoes escaping predation by shedding scales when grasped or touched, we propose a flexible escape skin by attaching artificial scales to a flexible film. Experiments demonstrate that the escape skin has significant effects in reducing escape force, escaping from harmful force environments, and resisting mechanical damage. Furthermore, we enabled active control of escape force and skin hardness by changing temperature, increasing the adaptability of the escape skin to the surrounding. Our study helps lay the foundation for engineering systems that depend on escape skin to improve robustness.

RESUMEN

Rigid-flexible-soft coupled robots are an important development direction of robotics, which face many theoretical and technical challenges in their design, manufacture, and modeling. Inspired by fishbones, we propose a novel cable-driven single-backbone continuum robot which has a compact structure, is lightweight, and has high dexterity. In contrast to the existing single-backbone continuum robots, the middle backbone of the continuum robot is serially formed by multiple cross-arranged bioinspired fishbone units. The proposed bioinspired fishbone unit, having good one-dimensional bending properties, is a special rigid-flexible-soft structure mainly made by multi-material 3D printing technology. The unique design and manufacture of the middle backbone provide the continuum robot with excellent constant curvature characteristics and reduce the coupling between different motion dimensions, laying a foundation for the continuum robot to have a more accurate theoretical model as well as regular and controllable deformation. Moreover, we build the forward and inverse kinematics model based on the geometric analysis method, and analyze its workspace. Further, the comparison between the experimental and theoretical results shows that the prediction errors of the kinematics model are within the desired 0.5 mm. Also, we establish the relation between the cable driving force of the bioinspired fishbone unit and its bending angle, which can provide guidance for the optimization of the continuum robot in the future. The application demos prove that the continuum robot has good dexterity and compliance, and can perform tasks such as obstacle crossing locomotion and narrow space transportation. This work provides new ideas for the bioinspired design and high-precision modeling of continuum robots.

RESUMEN

Long non-coding RNAs (lncRNAs) have been confirmed to be involved in epilepsy development. It has been reported that lncRNA ZFAS1 plays a vital regulatory role in epilepsy progression. Therefore, the role and molecular mechanism of ZFAS1 in epilepsy progression deserve further investigation. Mice status epilepticus (SE) model was constructed, and hippocampal neurons were isolated from mice hippocampus tissues. The expression of ZFAS1, miR-15a-5p and oxidative stress responsive 1 (OXSR1) were determined by quantitative real-time PCR. ELISA assay was used to detect the concentrations of inflammation factors. Cell viability and apoptosis were examined by MTT assay, EdU staining and flow cytometry. Western blot analysis was conducted to measure protein levels, and the productions of SOD and MDA were measured to assess cell oxidative stress. Dual-luciferase reporter assay and RIP assay were employed to validate the relationship between miR-15a-5p and ZFAS1 or OXSR1. LncRNA ZFAS1 was highly expressed in SE mice and SE-stimulated hippocampal neurons. Silenced ZFAS1 promoted viability, while inhibited inflammation, apoptosis and oxidative stress in SE-induced hippocampal neurons. MiR-15a-5p could be targeted by ZFAS1, and its inhibitor also reversed the suppressive effect of ZFAS1 knockdown on SE-induced hippocampal neurons injury. In addition, OXSR1 was a target of miR-15a-5p, and its silencing also could relieve SE-induced hippocampal neurons injury. OXSR1 overexpression reversed the inhibition effect of miR-15a-5p on SE-induced hippocampal neurons injury. Moreover, ZFAS1 positively regulated OXSR1 expression by sponging miR-15a-5p, thereby activating the NF-κB pathway. LncRNA ZFAS1 might contribute to the progression of epilepsy by regulating the miR-15a-5p/OXSR1/NF-κB pathway.

RESUMEN

Recently developed base editors provide a powerful tool for plant research and crop improvement. Although a number of different deaminases and Cas proteins have been used to improve base editors the editing efficiency, and editing window are still not optimal. Fusion of a non-sequence-specific single-stranded DNA-binding domain (DBD) from the human Rad51 protein between Cas9 nickase and the deaminase has been reported to dramatically increase the editing efficiency and expand the editing window of base editors in the mammalian cell lines and mouse embryos. We report the use of this strategy in rice, by fusing a rice codon-optimized human Rad51 DBD to the cytidine base editors AncBE4max, AncBE4max-NG, and evoFERNY. Our results show that the addition of Rad51 DBD did not increase editing efficiency in the major editing window but the editing range was expanded in all the three systems. Replacing the human Rad51 DBD with the rice Rad51 DBD homolog also expanded the editing window effectively.

RESUMEN

How to further improve the dexterity of continuum robots so that they can quickly change their structural size like flexible biological organs is a key challenge in the field of robotics. To tackle this dexterity challenge, this paper proposes a soft-rigid coupled bioinspired elephant trunk robot with variable diameter, which is enabled by combining a soft motion mechanism with a novel rigid variable-diameter mechanism (double pyramid deployable mechanism). The integration of these two mechanisms has produced three significant beneficial effects: (i) The coexistence of multi-degree-of-freedom motion capability and variable size function greatly improves the dexterity of the elephant trunk robot. (ii) The motion refinement can be improved by structural amplification, making up for the low resolution of soft actuators. (iii) Its stiffness can be increased by enlarging its diameter, while its reachable workspace can be increased by decreasing its diameter. Thus, the elephant trunk robot can optimize its performance when facing different tasks by opening and closing the rigid variable-diameter mechanism. Further, we established a kinematic model of the elephant trunk robot by the structure discretization method and the principle of mechanism equivalence, and experimentally verified its reasonableness. The demonstration experiments show that the elephant trunk robot has good flexibility. This work provides a new variable diameter configuration for continuum robots, and presents a method of how to analyze the kinematics of continuum mechanisms using rigid mechanism theory.

RESUMEN

In this study, a strain producing ß-glucanase and protease, identified as Bacillus velezensis Y1, was isolated from the manure of piglet. We attempted to produce ß-glucanase and protease after optimization of various process parameters with the submerged fermentation. The effects of each factor on producing ß-glucanase and protease were as follows: temperature > time > pH > loaded liquid volume. The properties of the ß-glucanase showed that the most suitable reaction temperature was 65 °C and pH was 6.0. However for protease optimum reaction temperature was 50 °C, and pH was 6.0. The amplified PCR fragments of ß-glucanase and protease were 1434 bp containing an open reading frame of 1413 bp encoding a protein with 444 amino acids and 1752 bp containing an open reading frame of 1521 bp encoding a protein with 506 amino acids, respectively. So, the study demonstrated a viable approach of using newly identified B. velezensis Y1 strain for the maximum yield of two industrially important enzymes.

RESUMEN

BACKGROUND: The excessive use of antibiotics in the livestock feed industry caused inevitable side effects of microbial resistance. Besides this residual antibiotics in animal-derived foodstuff imposed serious health problems for humans. So this study aimed to investigate the potential use of Bacillus velezensis to substitute antibiotics for poultry production. A total of 468, 49-week-old Hy-Line Brown chickens, were randomly divided into four groups the control group (regular diet), experiment group I (0.1% B. veleznesis), experiment group II (0.2% B. veleznesis), and antibiotic group (50 mg/kg flavomycin), with three replicates per group and trial period consisted on 42 days. RESULTS: The results showed that, compared with the control group, the average egg production rate and daily feed intake of experimental groups I and II increased significantly (P < 0.05), while the average egg weight was increased in experimental group II as compared to (I) (P < 0.01). The feed conversion ratio was decreased (P > 0.05) in group (II) Egg quality parameters such as yolk weight of the experimental group II was increased, but that of the antibiotic group and experiment group I was decreased, neither significant (P > 0.05). Moreover, the eggshell strength, yolk color, albumen height, and Haugh unit were significantly increased (P < 0.05). Compared with the control group, probiotic groups can increase the progesterone and motilin (P > 0.05) but decrease the secretin and cholecystokinin in the blood plasma (P > 0.05). CONCLUSIONS: This study suggested that B. velezensis can substitute in-feed-antibiotics and improved most of the study parameters significantly. Which suggested that B. velezensis has potential future application value to replace the feed antibiotics.

RESUMEN

We aimed to investigate the sex differences in the clinical characteristics and risk factors for adverse outcomes among elderly patients with atherosclerotic stroke. We recruited 942 consecutive patients with atherosclerotic stroke aged 75 years and older between January 2008 and December 2013 from Jiamusi University First Hospital, China. Stroke subtype, severity, risk factors, and outcomes (mortality, dependency, and recurrence) at 3 and 12 months after stroke were recorded and assessed. Mortality at 3 months after stroke was higher in men than in women. Stroke severity was an independent risk factor for mortality, dependency, and recurrence at 3 and 12 months after stroke in both men and women. However, the presence of total anterior circulation infarct and obesity protected against mortality at 3 months after stroke in men, while total anterior circulation infarct was a risk factor for dependency at 3 months after stroke in women. In women, positive associations were found between fasting plasma glucose level and mortality at 3 months after stroke and between hypertension, atrial fibrillation, and recurrence at 12 months after stroke. These findings suggest that it is crucial to control the primary risk factors individually by sex, especially regarding hypertension and atrial fibrillation management, to improve secondary prevention of stroke among the elderly and reduce the burden of stroke in China.
